ABSTRACT

Purpose To investigate the expression of the protein of Ezrin and AQP5 in the normal, dysplasia gastric and carcinoma of esophagogastirc junction tissues and to find out their relationship with the biologic behavior of the carcinoma of esophagogastirc junction in the southern area of Hebei Province. Methods Ezrin and AQP5 proteins were detected by immunohistochemical staining in 10 ca-ses of normal gastric tissue, 40 cases dysplasia and 165 cases of the carcinoma of esophagogastirc junction tissues, respectively. Re-sults All of the expression levels of Ezrin and AQP5 protein were increasing according to the order of normal-dysplsia-carcinoma. The expression rate of Ezrin was 10%, 45% and 72. 7%, while the rate of AQP5 with 20%, 32. 5% and 65. 5% in the normal, dysplasia and carcinoma subgroup. The expression rates of each protein were significantly different among the three groups (P<0. 05). The ex-pression of them in the subgroup of poorly-differentiated with serosa invasion and lymph nodes metastasis was significantly higher than the other subgroup of well-differentiated with non serosa invasion or lymph nodes metastasis (P<0. 05). There was a positive correla-tion between the expression of Ezrin and AQP5 proteins in the carcinoma of esophagogastirc junction (P<0. 05). Conclusions Over-expression of Ezrin and AQP5 is detected in the carcinoma of esophagogastirc junction, with positive relationships, which probably helps the invasiveness and metastasis in the carcinoma of esophagogastirc junction synergistically.

ABSTRACT

Objective To explore the changes of S-100 protein(S-100) levels in blood in newborn rats with bilirubin encephalopathy. Methods The model was established by administered bilirubin intraperitoneally(200 mg/kg) in newborn rats. The blood and brains samples were taken from 8 rats at the sequential time points of 6,12,24,48,72 and 96 h after the model established.Eight normal rats were used as the control. The dynamic changes of S-100 were detected by enzyme linked immunosorbent assay(ELISA) and immunohistochemical technique. Results S-100 significantly increased in blood of bilirubin encephalopathy newborn rats compared with controls(P

ABSTRACT

[Objective]To explore the expression of OPG/OPGL protein and its significance in rat model of trauma-induced osteonecrosis of the femoral head(ONFH). [Methods]Thirty-two SD rats about 6 months were divided randomly into experimental and control groups. The animal model of femoral head necrosis was established in 32 SD rats by removing round ligaments of femoral head.Animals were sacrificed at 1,2,4 and 6 weeks after operation, respectively. The specimens were examined through histological observation under light microscope. The other side with sham operation served as normal control group. The comparison of fat tissue with hematopoietic tissue in the cavity of bone marrow of femoral head were performed by CMIAS computer-assisted image and statistical analysis. The percentage of empty lacuna in the femoral heads was obtained.Immunohistochemistry was used to determine the expression of OPG/OPGL protein in ONFH.[Results]ONFH was confirmed in experimental group. The model in various stages was successfully duplicated.Compared with normal control group, the percentage of empty lacuna remarkable increase was found in experimental groups in different periods(P

ABSTRACT

The protein encoded by the Bcl-2 proto-oncogene has been shown to prolong cell su-rvival by preventing cell death(Apoptosis) induced by many insults including cancer therap-eutic drugs. Recently many researches have elucidated the bcl-2 expression in several hu-man solid cancers. However, there is still a few avaiable data to determine the role of Bcl-2 expression in the ovarian carcinogenesis and its prognostic significance in ovarian can-cers. Hence, we examined the expression of Bcl-2 in 68 ovarian epithelial cancers using immunohistochemistry and determined whether Bcl-2 expression has prognostic significance in the ovarian epithelial cancers. We found Bcl-2 expression(>5% positive cell) in 31 patients(40%). Bcl-2 expression were exclusively negative in the mucinous type of the ovarian epithelial cancer. Bcl-2 exp- ression was not correlated with tumor stage(stage I & II vs III & IV, p=0.63). The patients with Bcl-2 positivity had lower recurrence rate than the patients with negativity at the se- cond look operation(p0.05). These results sug- gest the Bcl-2 expression appears an early event in the ovarian carcinogenesis and has an inhibiting role in progression of ovarian tumor.

ABSTRACT

The distribution of GABA-containing neurons in the anterior horn of L_(4~5) segments of rat spinal cord and their relationship with somatic efferent were studied by combined method of HRP and immunocytochemistry and immunoelectron microscopic method. The results showed that under light microscope, the GABA-immunoreactive cell bodies and terminals were seen in all layers of the anterior horn including Rexed′s layer IX which located in the anterolateral part of the anterior horn. The GABA-positive neurons had round or triangle cell body with many processes and could be divided into two (large and medium) types. Under electron microscope, GABA-immunoreactive products appeared as small granular deposits located in perikarya, dendrite and axon. In axon terminals the immunoreactive products located at periphery of the synaptic vesicles and on the outer membrane of the mitochondria. The GABA-positive dendrites received symmetrical afferent synapses from GABA-positive or negative axon terminals. The combined method of HRP and immunocytochemistry showed that in Rexed′s layer IX there were HRP single labeled neurons, GABA single labeled neurons and HRP/GABA double labeled neurons. The double labeled neurons accounted for 79% of total HRP labeled cells. Above mentioned results first identified the neurons in Rexed′s layer IX of the anterior horn contain GABA which participate in somatic efferent and receive autoregulation from GABA-neurons and nonGABA-neurons at synaptic level.
